Studies on morphological plasticity are extremely important to plant breeding seeking to optimize genotype interactions with their environments. The present study evaluated 38 genotypes of Brachiaria ruziziensis cultivated during the rainy (October to March) and dry (April to September) seasons and examined variations in their anatomical characteristics. Fully expanded leaves were collected and fixed in FAA70 (formaldehyde: acetic acid: ethyl alcohol) for 72h and subsequently stored in 70% alcohol. An inexpensive alternative equipment for the plant material embedding in the paraffin under the vacuum BABT

The present work describes an equipment constructed using inexpensive material for embedding the plant material in the paraffin under the vacuum, using an oven and a vacuum pump. The equipment was tested using the samples of Rodriguezia venusta (Orchidaceae) buds embedded in paraffin, where half of the samples were submitted to the vacuum by the equipment during the embedding. The material was sectioned with a rotary microtome, obtaining full series of quality sections. The control was hard to section with the microtome, obtaining damaged sections due the air bubbles, making the ribbon formation difficult. These results proved the effectiveness of the equipment, making it a practical, inexpensive and more portable solution for newly established laboratories.

Anatomic compatibility of pear and quince trees grafted on Pyrus calleryana and Chaenomeles sinensis rootstocks PAB

Abstract: The objective of this work was to evaluate the graft growth and the anatomical characterization of pear (Pyrus communis x P. pyrifolia) and quince (Cydonia oblonga) cultivars grafted on Pyrus calleryana and Chaenomeles sinensis rootstocks. Sprouted graft percentages at 60 days and the length and diameter of the graft sproutings at 120 days were evaluated. Stem cuttings from the grafting region were performed in transversal and longitudinal sections, and the secondary xylem was separated for tissue comparison. The present work characterized and compared the anatomical structures of the leaves of Bactris gasipaes (Arecaceae) plants grown under different cultivation conditions (in vitro, ex vitro and in vivo) with the goal of identifying the origins of the difficulties encountered in acclimatizing micro-plants. The Quant program was used to determine leaf tissue thicknesses and areas, and histochemical tests were performed on leaf sections and analyzed using light microscopy. As lamina anatomy characteristics are relevant in circumscribing Arecaceae taxa, this work aimed to describe B. capitata lamina anatomy and compare it with B. odorata. O objetivo deste estudo foi conhecer a estrutura populacional e indicar usos para Guadua cf. superba, com base nas propriedades anatômicas do colmo (haste com nós e entrenós bem visíveis). A população estudada está localizada na Reserva Extrativista Chico Mendes, no Acre. The present work aims to demonstrate the stages of fabrication and use of a simple device to avoid the formation or fixation of precipitates from Sudan Black B solution on tissues. The device consists of four coverslip fragments attached to a histology slide, which serve as points of support for the histological slide under analysis. To work properly, the histology slide with the sections should be placed with the sections facing downwards the device. A small space between the device and the histology slide is thereby created by the height of the coverslip fragments. This study aimed at verifying the lead tolerance of water hyacinth and at looking at consequent anatomical and physiological modifications. Water hyacinth plants were grown on nutrient solutions with five different lead concentrations: 0.00, 0.50, 1.00, 2.00 and 4.00 mg L–1 by 20 days. Photosynthesis, transpiration, stomatal conductance and the Ci/Ca rate were measured at the end of 15 days of experiment. A study on the leaf anatomy of Cordiera sessilis (Rubiaceae), a native medicinal shrub from Brazilian Cerrado was carried out to contribute for species identification and provide support for future biochemical studies and medicinal application. Leaves were collected, fixed and processed by usual techniques, and studied by light and electron microscopy. Quantitative analyzes of stomata and trichomes were performed. Abstract: The objective of this work was to characterize morphologically elephant grass (Pennisetum purpureum) genotypes and to estimate their proportions of leaf blade tissues under grazing. Two tall varieties (Elephant B and IRI-381) and three short ones (Mott, Taiwan A-146 2.37, and Taiwan A-146 2.114) were evaluated under intermittent sheep grazing as to the following morphological characteristics: plant height, internode length, and leaf blade/culm ratio. Moreover, the proportions of the following leaf blade tissues were estimated: sclerenchyma, adaxial and abaxial epidermis, bulliform cells, vascular bundles, phloem, vascular sheath, xylem, and mesophyll. In specialized literature, reports on anatomy of miners in host plants are few in number. These agents trigger excavations, or paths, by consumption of plant inner tissues by larvae of several insects. The aim of this work was to investigate leaf miner occurrence in Commelina diffusa (a cosmopolitan plant) and Floscopa glabrata (an amphibious plant) using anatomical techniques. Extrafloral nectaries (EFNs) are structurally variable and widely spread among the angiosperms. The occurrence of EFNs in leaves of Pterodon polygalaeflorus Benth. and Pterodon pubescens Benth. (Fabaceae: Papilionoideae) were detected in adult specimens, at the time of production of new buds and flowers. The goals of the present study are to register the occurrence of the EFNs in P. pubescens and P. polygalaeflorus, and provide comparative data on the anatomical structures.
